6L6 is fat and round and obviously has more output. The 6V6 is sweeter sounding but has less output and sounds closer to an EL34.
It depends on what sound you are looking for and how much output power you need.
A 4-6V6 amp has a complex sound and can make about 30-40 watts output under normal operating parameters.4-6L6's can put out 100 watts or more.
The tubes are different animals that's for sure.
Crystal latice or vacuum, that is the question.

A minor clarification not all 6L's deliver the same amount of power...for instance, the 6L6GC has 30W and the 7581A has 35W 6L6WGB 19w maximum plate dissipation ratings....here are the NOS published values (Ppd & Vp):

19W, 360V = 6L6/6L6G/6L6GA/6L6GB
19W, 360V = 6L6WGB/5932
23W, 400V = 5881
25W, 500V = 7581
26W, 400V = 6L6WGB; Ps = 3.5W
30W, 500V = 6L6GC; Ps = 5.0W
35W, 500V = 7581A
35W, 600V = 7027A

FWIW..I think of the 6V (NOS) as the little brother of the 6L/5881 and the 6K6 as the little brother of the 6V..

Certainly a fatter sounding tube is more my preference, i use 6L6 ,s in my Traynor which sound awesome with my strat :D but pants with my PRS
Hopefully the Dumble build i,ve just started will cure my PRS blues

I owned a PRS Custom 24, Ten Top, Birds.. Lovely guitar it was the most seamless and playable guitar i've ever played and it stayed in tune amazingly well.

Unforunately it did sound a bit pants for what I paid.

Mines a early CE22 with a really nice flame top all in trans blue, sounds great through a boogie or almost anything else apart from the Traynor, i suspect that if i used the eq in my TC Nova i could improve things but its a great excuse to build a amp
